Comments: The HT-2 cell line was derived in 1979 by J. Watson at the University of Auckland from a culture of BALB/c spleen cells activated by sheep erythrocytes in the presence of IL-2.

The A5E clone was selected to provide a homogeneous population that is highly sensitive to IL-2.

The line is dependent on IL-2 for growth and can be used to measure IL-2 activity in a very rapid and sensitive assay.

The cells are commonly used for characterization of lymphokines and cytokines.

In addition to IL-2, substantial proliferation is seen in the presence of interleukin 4 (interleukin-4, IL-4, also known as BSF-1).

Propagation: ATCC complete growth medium: RPMI 1640 medium with 2 mM L-glutamine adjusted to contain 1.5 g/L sodium bicarbonate, 4.5 g/L glucose, 10 mM HEPES and 1.0 mM sodium pyruvate and supplemented with 0.05 mM 2-mercaptoethanol, 100-200 IU/ml IL-2, 90%; fetal bovine serum, 10%Alternatively, IL-2 in the form of Rat T-STIM factor with Con A (available from Becton Dickinson, Cat.# 354115) may be used at 10% by volume.

Subculturing: HT-2 clone A5E細胞Medium Renewal: Every 2 to 3 days

Cultures can be maintained by addition or replacement of fresh medium. Subcuture every two days at 3.5 X 10 exp4 viable cells/ml.
